Intended for use:
Canine Brucella Antibody Rapid Test is a lateral flow immunochromatographic assay for the qualitative detection of Canine Brucella Antibody (C.Brucella Ab) in dog’s serum, plasma, or whole blood.
Assay Time: 5-10 min
Sample: serum, plasma, or whole blood. (Will not require Blood Centrifugation)
Principle of the Test:
Canine Brucella Antibody Rapid Test is based on sandwich lateral flow immunochromatographic assay. The test device has a testing window for the observation of assay running and result reading. The testing window has an invisible T (test) zone and a C (control) zone before running the assay. When the treated sample was applied into the sample hole on the device, the liquid will laterally flow through the surface of the test strip and react with the pre-coated Brucella LPS antigens. If there are anti-Brucella antibodies in the specimen, a visible T line will appear. The C line should always appear after a sample is applied, which indicates a valid result. By this means, the Test can accurately indicate the presence of Canine Brucella antibodies in the specimen.
Test Procedure utilizing whole blood:
1- Collect dog’s fresh whole blood
2- Take out the test device from the foil pouch and place it horizontally.
3- using the provided dropper Immediately aspirate one drop of the fresh whole blood and add into the assay diluent.
4- Mix well for a few seconds.
5- Place 2-3 drops (approx 100-150μL) of assay buffer into the sample hole.
6- Interpret the result at 5-10 minutes. Result after 10 minutes is considered as invalid
Test procedure utilizing Serum:
1- Draw 1-1.5mL blood from your dog
2- let the blood tube in room temperature for 10 to 15 minutes for blood coagulation.
3- place the tube in the Centrifuge and spin for 5 to 7 minutes at 4000rpm or at higher speed If there is no centrifuge available, after collecting the blood, keep the blood tube (plain Tube) in a standing position for 8-10 hours at 2-8C (in the refrigerator) then aspirate 1 drop of the Serum and add into the assay diluent buffer and mix thoroughly.
4- Take out the Test strip from the foil pouch and place it horizontally.
5- using the provided dropper and aspirate one drop of the serum and add into the assay diluent buffer.
6- Mix well for a few seconds.
7- Place 2-3 drops (approx 100-150μL) of assay buffer into the sample hole.
8- Interpret the result at 5-10 minutes. Result after 10 minutes is considered as invalid
Interpretation of Test Result:
– Positive: The presence of both “C” band and “T” band, no matter “T” band is clear or vague.
– Negative: Only clear “C” band appears.
– Invalid: No colored band appears in “C” zone, no matter whether “T” band appears.
Storage:
– The kit can be stored at room temperature (2-30°C).
– DO NOT FREEZE. Do not store the test kit in direct sunlight.
